In simple words, serial data transfer might have many possible methods, know as protocols. Serial eproms don't have same protocol as that of a serial port of PC. Thats why, it is not possible directly.

However, you can use the parallel port of PC to directly program a serial eprom, since on parallel port, it is the user/programmer who can decide the protocol for data transfer.
