There is a lot of open source comm code available; check out http://freshmeat.net. There are also a lot of books covering programming serial ports, like Joe Campbell's "C Programmer's Guide to Serial Communications."
Then there are the Serial-HOWTO info sites. Remember that while a lot of this is for Linux, most Linux machines are PC-based so you've got the same hardware to initialize.
