No, the boot loader does not have to use interrupts.

But the main application may have to use interrupts. And if the boot loader is stored at the location where the processor looks for interrupt handlers for the application ISR, the boot loader must have a mechanism for forwarding the interrupt responses to the ISR in the application memory space.
