Use other unused line of the RS232 standar.
You can activate it in software in the PC side before starting the transfer.

Or only check the line to check the start bit (the code is shorter) and insert this code in the main loop of your device.

Why don't use a derivate with uart?
Or an external uart with I²C interface?
