As far as i remember having 16 BIT ADCs on 2 layer boards need very good Ground routing.
Are you really into the need of 16 bit resulotion ?If so, and you Don't have a good ground plane here are my suggestions(in case that you didn't already knew them) :
- If you need a sample each second Try to take samples in 1/100 second and equalize them.this will give you a better result.
- Use a RC filter in your Sensor connects to the ADC.
