Is there any specific thing that differentiate these two, with respect to functionality, or the purpose of their use, they way they are used.

Haven't I already explained that?

Are both same in functionality.

Definitely not.

Eg. Sometime we store data in buffer and some time in port.

It would be extremely unusual to think of a port as "storing" data - but, again, it depends very much on the context.

What is the use of storing data in port or in buffer.

Depends on which particular buffer and which particular port you're talking about!
