It is possible to make a 'universal' smartcard reader, but it takes some work. The difference between a synchronous (I2C, three wire) and asynchronous card (T=0, T=1) can be detected by the way in which they respond after a reset. The differences between any of these cards separately can be read from the ATR (or, in the case of a 'dumb' memory card, the lack of ATR). To make your life easier, I would suggest you take a look at Atmel's range of Smartcard Reader IC's. These are 8051 compatible controllers that have everything on board to make a complete smartcard reader. They also include a DC/DC converter, so 3V and 5V cards can both be handled.
