If you have the COMM port open in a DOS box under the likes of Win 2K or XP the port will be unavailable to Hyper terminal.

From what I know there is NO way to control the handshake lines directly from inside HyperTerm. I even checked the somewhat more robust HyperTerminal Private Edition that I bought from Hilgrave SOftware and it seems to have the same limitations.

So I think maybe you are stuck either making your own terminal emulator program or finding another one with source code wherein you can add the functionalit you require.
